Purification and Characterization of a Bacteriocin Produced by Lactobacillus lactis Isolated from Marine Environment

چکیده
Bacteriocin producing Lactobacillus lactis strain isolated from marine environment, showed broad range of antibacterial activity against some major food borne pathogens. Maximum bacteriocin production was observed at 30°C , pH 6.0 and 1.5% sodium chloride so lution. In addition of enzymes, "-amylase, DNase, RNase and lipase were slightly positive effect bacteriocin production. Proteinase K and pepsin were strongly inhibited bacteriocin production. Among detergents, Sodium dodecyl sulphate (SDS), Tween 80 and Tritone X-100 stimulated bacteriocin production and strongly inhibited by EDTA and urea. The bacteriocin has purified by ammonium sulphate precipitate and ion exchange (DEAE cellulose) chromatography. Biochemically it was pure protein moiety and the molecular weight was 94 kDa. The study revealed the possibility of using bacteriocin as a food preservative and the L. lactis strain as probiotic.

Background: Bacteriocins are small peptides which are ribosomally synthesized and have been shown to have wide range of antimicrobial activity. The aim of this study was to optimize the production of L. paracasei MG847589 bacteriocin. Furthermore, the potential antibacterial properties of the novel bacteriocins were characterized and evaluated against Staphylococcus aureus. Methods: The presen...
متن کاملEffect of Bacteriocin Produced by Lactobacillus casei with Probiotic Potential Isolated from Milk on the Ability of Biofilm Production by Streptococcus salivarius Obtained from Dental Plaque
Background and purpose: Dental plaque is a biofilm that is formed on the surface of the tooth. Carious lesions are caused by inappropriate ecological changes in microbial flora of the plaque biofilm. In this study, the effect of Lactobacillus casei bacteriocin, isolated from dairy products, was investigated on Streptococcus salivarius biofilm formation. Materials and methods: In this experimen...

ABSTRACT- Lactic acid bacteria are commonly found in the gastrointestinal tract of various endothermic animals and humans, in milk, dairy and seafood products and on some plant surfaces. The aim of this study was to investigate the inhibitory effect of isolated lactic acid bacteria from intestines of narrow-barred Spanish mackerel (Scomberomorus commerson) on Listeria innocua growth. Five strai...
متن کاملGenotypic and Phylogenic Analysis of Lactobacilli Producing Bacteriocin Isolated from Traditional Dairy Products and Food
Background & Objective: Lactic acid bacteria (LAB) are a group of Gram-positive, non-spore forming, cocci or rod shaped, catalase negative organisms, considered as Generally Recognized as Safe (GRAS) organisms. These bacteria are used for thousands of years for production of fermented foods because of their ability to produce desirable changes in taste, flavor and texture. Different antimicrobi...
متن کاملPurification and genetic characterization of plantaricin NC8, a novel coculture-inducible two-peptide bacteriocin from Lactobacillus plantarum NC8.
A new, coculture-inducible two-peptide bacteriocin named plantaricin NC8 (PLNC8) was isolated from Lactobacillus plantarum NC8 cultures which had been induced with Lactococcus lactis MG1363 or Pediococcus pentosaceus FBB63. This bacteriocin consists of two distinct peptides, named alpha and beta, which were separated by C(2)-C(18) reverse-phase chromatography and whose complementary action is n...
